There are two isoforms of 11β-hydroxysteroid dehydrogenase: 11β-HSD1 and 11β-HSD2. 11β-HSD1 is expressed at high levels in the liver with a relatively low affinity for steroids, catalyzing both dehydrogenation and the reverse reductase reaction in a NADP+/NADPH-dependent manner. 11β-HSD2 is the enzyme that in the presence of NAD+ converts cortisol to cortisone, and is highly expressed in sodium-transporting epithelia, placenta, pancreas, and thyroid. The enzyme activity of 11β-HSD2 thus protects the nonselective mineralocorticoid receptor from being occupied by cortisol, which can cause hypertension. Mutations in the 11β-HSD2 gene cause juvenile hypertensive syndrome, aka apparent mineralocorticoid excess (AME). Acquired hypertension can arise from ingestion of licorice, which contains a competitive inhibitor of 11β-HSD2. Our 11β-HSD2 enzyme assay kit can be used to measure the activity of the enzyme present in cell/tissue crude lysates and serum. The colorimetric assay is based on the reduction of the tetrazolium salt INT in a NAD-coupled enzymatic reaction, yielding a water-soluble formazan product, which exhibits an absorption maximum at 492 nm. The intensity of the cherry-red color formed is increased in the presence of increased 11β-HSD2 activity. Kit components are stable for many years if handled and stored properly.
