Alizarin red S is a pigment originally extracted from the roots of the madder plant. The red dye was widely used to stain calcium-containing tissues such as bone and teeth in pre-clinical studies and was later chemically synthesized in 1869. Unlike von Kossa staining, which stains phosphate, Alizarin red stains primarily calcium, and both histological stains remain popular for identification of mineralized tissues in biomedical research. However, Alizarin red also binds other divalent cations such as magnesium and manganese.
