Alkaline phosphatase (ALP or ALK) possesses a hydrolytic activity removing phosphate groups from protein and non-protein molecules. ALP is present in all mammalian tissues, and multiple isozymes are differentially distributed in the body. While abnormal ALP levels can signal a wide range of clinical conditions, the primary importance of measuring ALP is diagnosis of bone or liver disease, although an elevated ALP usually requires other tests to determine the origin of the pathology. ALP elevation also serves as an osteogenic differentiation marker. The enzyme assay kit is based on ALP-mediated conversion of p-nitrophenol phosphate (PNPP) to nitrophenol in an alkaline buffer. The product nitrophenol exhibits an absorption maximum at 405-415 nm, allowing for sensitive detection of ALP activity in crude cell/tissue extracts and serum/plasma. The assay is designed for the 96-well plate format, but can be scaled up if desired. Repeated freeze-thaw cycles of the assay solution should be avoided.
