Enhanced Chemiluminescence Detection (HRP/luminol)

Product Description:
Detection by chemiluminescence has now become the method of choice for fast and sensitive assays of enzyme
conjugates on blots. While phosphate-containing dioxetane compounds are widely used as chemiluminescent substrates for alkaline phosphatase
conjugates, the compound luminol and its derivatives (diacylhydrazides) are the chemiluminescent substrates of choice for horse radish
peroxidase (HRP) conjugates (Methods Enzymol. 133:331,1986). HRP catalyzes the oxidation of luminol in the presence of hydrogen peroxide,
during which an excited intermediate is formed and subsequently decays to the ground state by emitting light. Light emission can be enhanced by
various enhancers acting as radical transmitters between the formed oxygen radical and luminol, thus increasing the detection sensitivity of the
HRP-luminol reaction significantly. The combined use of luminol and an enhancer system is thus the basis of ECL application. Our ECL HRP
Substrate System comprises two separate solutions: Peroxide Solution and Luminol Solution. The product is for research use only.
