Product Description: Ketone bodies, like short-chain fatty acids, are water-soluble, and include acetoacetate (AcAc), beta-hydroxybutyrate
(BHB), and acetone. If dietary carbohydrate is insufficient or is not properly utilized due to insulin deficiency, the level of hepatic oxaloacetate will
not be adequate to ensure oxidation through the TCA cycle of all the acetyl-CoA generated from fatty acid oxidation. A low level of AcAc and BHB
are always present in the blood, and their levels increase during fasting and prolonged exercise. Ketogenesis also occurs as a result of a
high-fat, low-carb diet. Ketone bodies released from the liver are absorbed by the brain and peripheral tissues. Ketoacidosis occurs if an
abnormally high amount of ketone bodies, which are carboxylic acids, is present in the circulation. This situation can be seen with diabetic
ketoacidosis (DKA), which can lead to coma or even death if ignored. When blood ketone body levels increase beyond ~0.2 mM, excretion
increases and ketone bodies become measurable in the urine. Since BHB represents ~75% of the ketone bodies in the blood, measurement of
BHB provides reliable information for patient management and DKA monitoring. Our BHB Assay Kit is based on the reduction of the tetrazolium
salt INT in a NADH-coupled enzymatic reaction to formazan, which exhibits an absorption maximum at 492 nm. The assay kit can measure the
concentration of BHB (sensitivity ~20uM) in the cell and released to the culture medium, circulation, or urine in a quantitative manner. The
non-radioactive assay allows measurements of serum, plasma, and urine samples to be completed in 30-60 min. The assay solution is stable for
at least one year if stored and handled properly.
